In the anime, the Pokéblock Case was first featured in Now That's Flower Power!, in which May received one from Mr. Big, who said that every Pokémon Coordinator should have one. Being a former Coordinator himself, Mr. Big talked to May about the importance of planning a Pokémon's diet and explained that different Berries can be used to produce Pokéblocks.

In Lessons in Lilycove, Kelly used two Pokéblock Cases during the Appeals Round of the Lilycove City Pokémon Contest. She released a large amount of green-colored Pokéblocks for her Grumpig to manipulate through the use of Psychic before eating them all. The performance was well received by the panel of judges and Kelly was among the eight contestants moving on to the Battle Round.

Drew, one of May's rivals, was revealed to own a Pokéblock Case in The Unbeatable Lightness of Seeing!, in which he was shown feeding his Roselia a few Pokéblocks.

In Reuniting for the First Time!, Chloe received a Pokéblock Case from Lisia after she had scouted her for a potential performance at the Lilycove City Contest Spectacular.
