Tapu Koko first makes its debut in the original Sun and Moon titles, where it presents itself to the [[Player character|player]] and [[Lillie]] at the beginning of the game. It contains mostly the same role in both titles but species may differ.

At the beginning of both titles, the player will find an uneasy Lillie, attempting to save her {{p|Cosmog}}, [[Nebby]], from a flock of wild {{p|Spearow}} on an unsteady [[Mahalo Trail|blank bridge]]. After the player's attempt to guard Nebby, the Legendary Pokémon uses its power to collapse the bridge, causing the two to nearly fall into the rushing river below. Tapu Koko's lightning quick speed not only knocks the Spearow away from the player and Nebby, but also grabs the two before hitting the river and rushes them towards the entrance of Mahalo Trail where Lillie first attempted to cross. The player, Lillie and Nebby all exchange a quick glance with the guardian deity that had appeared before them. Tapu Koko once again, flies away at quick pace, leaving the group in disbelief. Lillie notices a [[Sparkling Stone]] on the ground and offers it to the player before they make their way back to [[Iki Town]]. This Sparkling Stone is then given to [[Hala]] by the player and is turned into either a [[Z-Ring]] or a [[Z-Power Ring]] by the time they pick it up the following day. The same day where the player receives this item is also the same day where they and Hau begin their [[island challenge]], and a festival to commemorate their inauguration is held. A [[Pokémon battle]] between the two Trainers is held to honor Tapu Koko, who is very pleased by the results of the battle, starting their island challenge. After the player makes their trip around Melemele Island on their island challenge, they will be met with a grand trial and a battle against the island kahuna, Hala. This grand trial is also held in praise of Tapu Koko. Tapu Koko is happy with the results of the player's efforts against Hala and lets out a roar. Hala explains that Tapu Koko may be excited to battle the player once they gain the strength to do so.

In Pokémon Ultra Sun and Ultra Moon, Tapu Koko appears after becoming {{pkmn|Champion}}. If defeated or run from, it respawns after the player [[Pokémon League (Alola)/Title Defense|defends their title]]. If Tapu Koko is the first of the four [[Guardian deities]] fought, it will drop a [[Tapunium Z]] after the battle.
