Unlike in Generation I, Guillotine can affect a Pokémon whose Speed stat is greater than the user's current Speed stat. However, it cannot affect a Pokémon of a higher level. The accuracy of Guillotine depends on the level of the user and the level of the target and is worked out with the formula: Accuracy = ((level of user - level of target) + 30)%. Guillotine ignores all changes to accuracy and evasion stats.

In Generation VI, through the use of the ability {{a|Aerilate}} if a Pokémon has it, Guillotine is made a Flying-type move. For this reason, it can KO Ghost-types without the need of {{m|Foresight}}, {{m|Odor Sleuth}}, or the ability {{a|Scrappy}}. Guillotine can KO Shedinja, as its' {{a|Wonder Guard}} will not protect it from Guillotine while the Pokémon using it has Aerilate.
In Generation VI, through the use of Normal-type changing abilities such as {{a|Aerilate}}, {{a|Pixilate}}, or {{a|Refrigerate}}, Guillotine is made a Flying, Ice, or Fairy-type move. For this reason, it can KO Ghost-types without the need of {{m|Foresight}}, {{m|Odor Sleuth}}, or the ability {{a|Scrappy}}. Guillotine can also KO Shedinja, but only if the Pokémon has Aerilate.
