In the [[Detective Pikachu (movie)|movie]], R is a gaseous chemical that causes exposed Pokémon to lose control. Unlike the games, the Pokémon's eyes turn purple and the Pokemon do not appear to tire after the effects wear off. The substance was created by [[Dr. Ann Laurent]] at the [[Pokémon Comprehensive Laboratory|PCL]] under the direction of [[Howard Clifford]]. In an attempt to find a cure of his degenerative condition, Howard believed that such issues could be solved if humans could evolve into better versions of themselves using the R substance.

[[Tim Goodman|Tim]] first comes across R when searching his father's apartment. He accidentally releases the fumes, causing a group of nearby {{p|Aipom}} to go berserk and attack Tim and {{OBP|Detective Pikachu|character}}. When Tim and Pikachu go to find [[Lucy Stevens|Lucy]], a CNM employee investigating [[Harry Goodman]]'s death, she shows them several vials of R that she has come across during her investigation.  

Once the pair track down a {{p|Mr. Mime}} at Ryme Wharf, it reveals that the R substance is being distributed at the "Roundhouse", an underground facility where illegal Pokémon battles are held. At the Roundhouse, {{OBP|Sebastian|Detective Pikachu}}'s {{p|Charizard}} battles Pikachu after Sebastian uses R on his Charizard. During the battle, Sebastian becomes knocked down by Charizard, causing the vials of R inside his coat to break and the gas to spread over the entire room. As the Pokémon fly into a frenzy, Sebastian tells Tim that R was being given out by someone he knows only as "the doctor."

Lucy suspects that the work of Dr. Laurent is connected to R and Harry's death. The three head off to the now abandoned PCL and discover that Laurent created a neural link headband for human use and had been extracting Mewtwo's DNA to create the R serum. Misled by Howard that Roger is behind the distribution of R, Tim, Lucy and Pikachu rush back to Ryme City. Tim goes to Howard's office, only to discover Howard wearing the neural link headband Laurent created. Howard reveals that he was the mastermind of the plot and that he plans to merge his mind with Mewtwo's body, although this only works if the Pokémon has lost control. Howard then releases the R substance to a captured Mewtwo, making it go berserk, and then merges his mind to Mewtwo using the neural link headband. Now possessing Mewtwo's body and its powers, Howard flies off into the city and ruptures the parade balloons which are filled with R. Using Mewtwo's unique ability, Howard transfers the minds of humans into all affected Pokémon in the city.
