 The fact that May's Beautifly was the only one of her Pokémon which enjoyed her first [[Pokéblock]]s which she made (that were LiteBlue) suggests that May's Beautifly enjoys bitter Pokéblock. This therefore suggests that its nature is either Calm, Careful, Sassy or Gentle. Though, judging from its calm and mellow demeanor which it exhibits as a Beautifly, this may mean that May's Beautifly has a Calm [[nature]].
